O papa leu o texto da missa em português, mas fez a homilia em espanhol. Embora os participantes fossem, na maioria, brasileiros, havia uma delegação de 80 peregrinos e três bispos das Ilhas Canárias, onde Anchieta nasceu na cidade de São Cristóvão da Laguna. Entre os religiosos brasileiros estavam presentes à missa d. Raymundo Damasceno Assis, d. Odilo Scherer, d. João Braz de Aziz, d. Cláudio Hummes e d. José Freire Falcão.
Cerca de 200 padres, mais da metade deles alunos do Colégio Pio Brasileiro, ocuparam as alas centrais da igreja. O superior geral dos jesuítas, padre Adolfo Nicolàs, sentou-se na primeira fila, ao lado do postulador das causas de canonização da Companhia de Jesus, padre Anton Witwoer, e do vice-postulador, padre César Augusto dos Santos. Eles presentearam o papa com uma relíquia de Anchieta, um pedaço do fêmur que se encontra no Pátio de Colégio, em São Paulo.
